To keep retain heat and humidity better, I just closed off the top of my enclosure with a sheet of plexiglass and sawed a hole out for the light fixture. Is it necessary to drill air holes for ventilation? I don't want too much heat and humidity to escape, but I dont want to suffocate him either...

No air holes.

He won't suffocate. You'll access the enclosure for food and water and air exchange will take place.

You'll be just fine :)

Ditto. Besides the holes you did drill, unless a perfect fit and caulked to seal it air tight, you will get air exchange through that too.
